Semi - Vitro Culture of Oenothera Pollen as Influenced by Calcium Application


Abstract
Convenient plants for the present type of experiment, Oenothera organensis and O. biennis, were used to obtain their stylar segments with stigmatic surface. After pollination the segments were inserted in an artificial medium containing 10% sucrose, 100 §·/§¤ boric acid and 1% agar. Whenever experimental treatment was given, soluble form of Ca was used. Ca ions in Allen cultural media remarkably enhance pollen germination and tube growth when cultured in vitro. This was also true if the pollen were cultured in semi-vitro of stylar segment.
This suggested strongly that pollen growth in styles under the natural conditions is also effectively exerted when optimal level of Ca ions is present.
